Early morning urine is the gold standard, the kit instructions say the same. I set an alarm and tested right after waking. Also test only after your missed period for best accuracy.
First thing in the morning, before drinking water, that's when the hormone is most concentrated. I tested afternoon once and got a faint line, morning gave a clear one. So always go for first morning urine.
