Best on the day your period is due or just after, using first morning urine. Testing before that often shows nothing even when you are pregnant. I always waited to avoid the false negative heartbreak.
Home kits are reliable around the missed period. If you really want an earlier answer, a blood beta hCG at the lab can detect a few days sooner, which is what my doctor suggested. But for most, waiting for the missed period is simplest.
